cell counting

Denise Dagnino
Hi

I am new to ImageJ. I am trying to count phytoplankton in an
hematocytometer. The cells, no matter how much I wait, do not sediment.
Further I can clearly notice that as time passes the volume rather quickly
decreases under the slide and this wolud result in wrong counting.

What I did was to take pictures in several planes. I would like manipulate
each of them so as to only observethe cells in focus and then stack all the
different focus plains and then count.

Any suggestions on strings of commands I should execute?
Thanks for any help.

Denise

Re: cell counting

G. Esteban Fernandez
Open all the planes and do Image > Stack > Images to Stack to collect them
into one window (called a "z-stack").

Then do Image > Stack > Z Project. Choose either Min or Max projection
depending on the type of images you took.

There are also plugins for "extended depth field" you can search for and
run on your z-stack.
